Of all of the comics ever published...which book would you recommend to a newcomer?

It's a tough call. I think I would try and see what kind of movies, TV shows, and books they are into. One of the best things in comics now right is that there are a TON of non-super-hero monthlies and OGNs that are offer lots of diversity.
Yeah, it really depends on what they're into.

Very good point Woodside. I would probably impose my will upon the newcomer and give them a copy of Avengers # 56...idk.

To showcase what the medium can really accomplish, I'd recommend Fables by Willingham and Buckingham.
It features characters known to almost everyone, fantastic art, features strong female characters that aren't trivialized or overly sexualized, and an outstanding storyline.
If they like that, then show the newcomer the different genres within comics so that they may pick what to read next.

As woodside rightly says, it would depend on the person involved. For some, Maus or Sandman would be great intros to comics. For others, Zot! or Concrete. Maybe Age of Bronze. Or Lee-Kirby FF circa #40-70. There is a lot of choice of good stuff, maybe the best thing would be to take them to a large comic book store and let them browse till they find something which catches their eye.
